Yersinia enterocolitica Typing by Restriction Enzyme Analysis of Plasmid DNA

Abstract
Forty-one Yersinia entrocolitica strains were collected in Korea and from abroad. Plasmid patterms of them were investigated to evaluate the differences of origin. Out of 41Y. enterocolitica strains, 14(34.2%) strains harbored one plasmid,
1(2.4%)
strain harbored two plasmids, while 26(63.4%0 strains were plasmid free. The sizes of plasmid harbored were 40-50MD for one plasmid, 7-8MD and 3-4MD for two plasmids, respectively. Fifteen strains which harbored plasmids were analyzed by applying
restriction enzyme(EcoR I, BamH I, Hind¥²) to distinguish restricted patterns. Restriction enzyme digestion patterns(REDP) proved that strains harboring plasmids were divided into 5 different types. Each type had several bands which were
distinguishable
from each other. The strains which belonged to the same REDP type had the indentical serotype. It may suggest that there is a relationship between REDP type and serotype b y these results. This technique could be used to evaluate the
epidemiologic
relationship between patients' samples and transmission vehicles, food which might be consjmed by patients, when a large outbreak of yersiniosis happes.
